DescriptionAtrochrysone carboxylic acid is a polyketide, a class of secondary metabolites characterized by their complex structures formed through the repetitive condensation of acetyl and malonyl-CoA units. It is synthesized by specific polyketide synthases, notably the atrochrysone carboxylic acid synthase (ACAS), which is a nonreducing, iterative type I polyketide synthase lacking a thioesterase domain (PMID:19549600 ). The synthesis pathway involves the condensation of malonyl-CoA to form the core structure of atrochrysone carboxylic acid, which can subsequently be hydrolyzed by the atrochrysone carboxyl ACP thioesterase (ACTE) to release related compounds like atrochrysone and endocrocin (PMID:19549600 ). Notably, CoPKS1 is known to produce atrochrysone carboxylic acid predominantly, while CoPKS4 exhibits dual activity, generating both hepta- and octaketides (PMID:36507600 ). Despite its biosynthetic pathway being elucidated, recent genomic analyses have not identified additional synthases for atrochrysone carboxylic acid or other oligocyclic metabolites (PMID:35218274 ). The heterologous production of this compound has been successfully achieved in Aspergillus oryzae, enhancing the supply of malonyl-CoA (PMID:31776294 ).
