| Description | Siamycin is a lasso peptide, a class of ribosomally synthesized and post-translationally modified peptides characterized by their unique topological structure, which includes a loop and a tail that is threaded through the loop. Chemically, siamycin-I has been shown to interact specifically with lipid II, a crucial component of bacterial cell wall biosynthesis, effectively targeting the Gram-positive cell surface and inhibiting peptidoglycan synthesis (PMID:31026131 ). This interaction disrupts the integrity of the bacterial cell wall, leading to cell lysis and death, particularly in vancomycin-resistant pathogens (PMID:40034853 ). Siamycin's mechanism of action is further underscored by its ability to restore vancomycin susceptibility in resistant strains of Enterococcus and Staphylococcus aureus, highlighting its potential therapeutic applications (PMID:40034853 ). Additionally, siamycin is associated with unique biosynthetic gene clusters that may encode novel compounds with significant biological activity, including cancer cell growth inhibition (PMID:39417076 ).
